Histidine is the only proteinogenic amino acid with a side-chain pKa in the physiological range — the imidazole group sits near pK 5.97 (Merck reports pK1' 1.78, pK2' 5.97, pK3' 8.97 for the free base). That is precisely why histidine is used as a buffer in biologics formulation, and it is also why a histidine addition will buffer your own product around pH 6 whether you wanted it to or not.
This is a monohydrate salt, so three different weights exist for the same material: the hydrated salt (209.63), the anhydrous salt (191.62) and the free base (155.15). Free histidine is 74.0% of the hydrated salt. Every potency calculation must state which basis it is on.
The hydrochloride form is chosen over the free base for solubility and handling — Merck describes the monohydrochloride as fairly soluble in water and insoluble in alcohol and ether, and the free base is only 45.6 g/L in water at 25 °C.
Water of crystallisation makes loss-on-drying a specification parameter rather than a purity check. A 'dry' lot that has lost its hydrate water will over-assay on the salt basis and under-deliver on a histidine-content basis.
In a beta-alanine stack, histidine is the co-substrate of the carnosine synthesis pathway, so the pairing is common commercially — but the two ingredients have very different solubility and taste profiles and should be evaluated separately in the flavour system.
