Histidine is the only proteinogenic amino acid with a side-chain pKa in the physiological range — the imidazole group sits near pK 5.97 (Merck reports pK1' 1.78, pK2' 5.97, pK3' 8.97 for the free base). That is precisely why histidine is used as a buffer in biologics formulation, and it is also why a histidine addition will buffer your own product around pH 6 whether you wanted it to or not.

This is a monohydrate salt, so three different weights exist for the same material: the hydrated salt (209.63), the anhydrous salt (191.62) and the free base (155.15). Free histidine is 74.0% of the hydrated salt. Every potency calculation must state which basis it is on.

The hydrochloride form is chosen over the free base for solubility and handling — Merck describes the monohydrochloride as fairly soluble in water and insoluble in alcohol and ether, and the free base is only 45.6 g/L in water at 25 °C.

Water of crystallisation makes loss-on-drying a specification parameter rather than a purity check. A 'dry' lot that has lost its hydrate water will over-assay on the salt basis and under-deliver on a histidine-content basis.

In a beta-alanine stack, histidine is the co-substrate of the carnosine synthesis pathway, so the pairing is common commercially — but the two ingredients have very different solubility and taste profiles and should be evaluated separately in the flavour system.

01The listing says 83% L-histidine. Is that right?
That figure needs checking and we would rather flag it than let you build a label on it. For L-histidine hydrochloride monohydrate, free histidine cannot exceed 74.0% of the salt mass (155.15/209.63) — even the anhydrous salt is only 81.0%. The 83% almost certainly refers to an assay of the salt, not to histidine content. Ask us for the manufacturer's specification sheet and we will get the basis stated unambiguously before you order.
02Do I need the hydrochloride or the free base?
Take the hydrochloride when you need solubility and easy handling; take the free base when you want maximum histidine per kilogram and can accept lower water solubility (45.6 g/L at 25 °C). For buffer work the choice is usually driven by the pH you are targeting and whether you can tolerate chloride.
03Why does histidine change my product's pH more than other amino acids?
Because it is the one amino acid whose side chain buffers in the range you care about — the imidazole pK is around 6. That is the whole reason histidine is used as a formulation buffer for biologics. Expect it to pull your system toward pH 6 and plan your buffer accordingly.
04How should the water content be specified?
As a range, not as a maximum. It is a monohydrate: theoretical water is 8.6%. A lot that has dried out will read high on a salt-basis assay and low on histidine content. Ask for Karl Fischer water on the COA.
05What is the MOQ for L-Histidine HCl Monohydrate?
